The Role of PPAR-γ and Its Interaction with COX-2 in Pancreatic Cancer
نویسنده
چکیده
In recent years, the study of the peroxisome proliferators activated receptor gamma (PPAR-gamma) as a potential target for cancer prevention and therapy has gained a strong interest. However, the overall biological significance of PPAR-gamma in cancer development and progression is still controversial. While many reports documented antiproliferative effects in human cancer cell and animal models, several studies demonstrating potential tumor promoting actions of PPAR-gamma ligands raised considerable concerns about the role of PPAR-gamma in human cancers. Controversy also exists about the role of PPAR-gamma in human pancreatic cancers. The current review summarizes the data about PPAR-gamma in pancreatic cancer and highlights the biologically relevant interactions between the cyclooxygenase and PPAR system.

15-Deoxy-Δ12,14-Prostaglandin J2 Protects PC12 cells from LPS-Induced Cell Death Through Nrf2 pathway in PPAR-γ Dependent Manner
Introduction: The inflammatory response requires a coordinated integration of various signaling pathway including cyclooxygenase (COX). COX catalyzes the formation of prostaglandins from arachidonic acid. Among prostaglandins, 15-Deoxy-D12, 14-prostaglandin J2 (15d-PGJ2), an endogenous ligand of Peroxisome proliferator-activated receptor-gamma (PPAR-γ), has been demonstrated to have anti-inflam...
متن کاملCompare the Effect of Eicosapentaenoic Acid and Oxidized Low-Density Lipoprotein on the Expression of CD36 and Peroxisome Proliferator-Activated Receptor Gamma
Background: There is evidence that CD36 promotes foam cell formation through internalizing oxidized LDL (ox-LDL) into macrophages therefore, it plays a key role in pathogenesis of atherosclerosis. In addition, CD36 expression seems to be mediated by nuclear receptor peroxisome proliferator-activated receptor gamma (PPAR-γ). The aim of the present study was to evaluate and compare the effect of ...
متن کاملSynergistic Antiproliferative Effects of Combined γ-Tocotrienol and PPARγ Antagonist Treatment Are Mediated through PPARγ-Independent Mechanisms in Breast Cancer Cells
Previous findings showed that the anticancer effects of combined γ -tocotrienol and peroxisome proliferator activated receptor γ (PPAR γ ) antagonist treatment caused a large reduction in PPAR γ expression. However, other studies suggest that the antiproliferative effects of γ -tocotrienol and/or PPAR γ antagonists are mediated, at least in part, through PPAR γ -independent mechanism(s). Studie...
متن کاملPioglitazone inhibits the proliferation and metastasis of human pancreatic cancer cells
Proliferator-activated receptor-γ (PPAR-γ) is a nuclear receptor that acts as a transcription factor in several types of tissue. PPAR-γ ligands are known to inhibit numerous cancer cell processes, including pancreatic cancer cell proliferation through terminal differentiation. Previous studies concerning the inhibitory effect of PPAR-γ ligands derived from thiazolidinediones (TZDs) on the metas...
متن کاملRole of Cyclooxygenese-2 (COX-2) Expression in Breast Cancer Differentiation and Its Relationship with Hormone Receptors Status
Background and Objectives: Cyclooxygenese-2 (COX-2) expression in breast cancer and its correlation with tumor prognosis is unclear. We investigated the incidence of COX-2 expression in patients and assessed interactions between COX-2 and clinical features of cancer and expression of HER2/neu, estrogen receptor (ER), and progesterone receptor (PR). Methods: COX-2 expression was investigated by...
